In this show, Marisa takes viewers on a journey across different regions of the world to taste and learn about various wines. The show begins its global tour in Portugal, where cork oak trees are prevalent, and the country is renowned for producing excellent ports and red wines. Marisa's excursions in Portugal take her to vineyards, wineries, and wine bars to taste wines such as Port, Douro, and Vinho Verde. She also learns about the country's wine history, methods of production, and culture.

From Portugal, Marisa and her team travel to France, the world's most famous wine country. The French leg of the journey takes the viewer to Burgundy, Champagne, and Bordeaux. In each area, Marisa explores winemaking techniques, grape varietals, terroir, and the unique wine culture of the region. She also attends wine festivals and trains with wine experts to gain insights into the art of wine tasting.

Following France, the series takes a stunning detour to Argentina, one of the world's most compelling wine producers, located in the southern hemisphere. Argentina has become famous for its Malbec wine, and Marisa takes a deep dive into the Malbec grape and its effect on the region's wine industry. She also learns about the history of Argentinean wine production, the ecological challenges facing the country's vineyards and the efforts made to promote sustainable wines.

The Wine Passport series also covers North America's famous wine regions, such as California's Napa Valley, Sonoma County, and Oregon's Willamette Valley. The North American episodes cover topics such as wine tourism, wine-making methods, flavor complexities, vineyard management, and the impact of climate change on wine production.

In addition to the above regions, the show also explores other unique wine-producing countries such as Georgia and Japan, where traditional winemaking techniques designed to work with the specific climate of the region have been in use for centuries. Marisa is intrigued by the use of clay pots to ferment wine in Georgia and also discovers Japanese wine made from the Koshu grape, a unique grape variety with an interesting flavor profile.
